Abstract :
In general terms, eukaryotic genes are mostly made of non-coding introns, separated by relatively much smaller exons. During RNA transcription a large complex assembles on the nascent transcript and precisely splices together the scattered coding exons. This process, known as pre-mRNA splicing, relies on the precise identification of intron-exon boundaries, which in the human genome contain apparently little information and therefore are difficult to predict. Furthermore, this identification can be regulated by many molecular strategies that are still poorly understood. Indeed, from a given gene often a precise population of transcripts is produced, and it defines the real expression of that gene. Consistent with this, alterations in the mRNA proportions in this set are deleterious for the organism. Thus, to fully decipher our genetic potential these questions will have first to be solved. This is also key to properly address diverse genetic diseases, ranging from neuropathies to distrophies and cancer, that are originated or promoted by mutations affecting pre-mRNA splicing. During my talk I will discuss these points and describe how we are working on them using two apparently dissimilar model systems: yeast and human.
